Product Description
Mouse embryonic stem cells (ES cells) are pluripotent cells derived from the inner cell
mass of day 3.5 blastocysts. They can be maintained in vitro for extended periods without
loss of their capacity to contribute to all cell lineages when re-implanted back into a
blastocyst. The pluripotency of mouse ES cells, combined with their ease of genetic
manipulation and selection, has revolutionized gene analysis studies in vivo via the
generation of transgenic, chimeric and knockout mice. Culture of mouse iPS cells and ES
cells requires the use of carefully pre-screened cell culture reagents to ensure retention of
pluripotency. All ES-Cult reagents are pre-screened to ensure the maintenance of mouse
ES cells. Mouse Embryonic Stem Cell Medium (MESCMS) is a sterile, liquid medium
which contains essential and non-essential amino acids, vitamins, organic and inorganic
compounds, growth factors, trace minerals. The medium is HEPES and bicarbonate
buffered and has a pH of 7.4 when equilibrated in an incubator with an atmosphere of 5%
CO2/95% air.
Components
MESCMS consists of 75 ml of fetal bovine serum (FBS, Cat. No. 0500), 20 ml of
embryonic stem cell supplement (ESC-S, finial concentration: 1% penicillin and
streptomycin, 2 mM L-glutamine, 0.1 mM non-essential amino acids and 0.1 mM
2-mercaptoethanol), 5 ml of leukemia inhibitory factor solution (LIF Cat. No.20150010.
Product Use
MESCMS is for research use only. It is not approved for human or animal use, or for
application in in vitro diagnostic procedures.
Storage
Store the FBS, ESC-S and the LIF solution at -20℃. Protect from light.

Prepare for use
Thaw FBS, ESC-S and LIF solution at 37℃. Gently tilt the tube several times during
thawing to help the contents dissolve. Make sure the contents of the supplement are
completely dissolved into solution before adding to the medium. Rinse the bottle and
tubes with 70% ethanol, and then wipe to remove excess. Remove the cap, being careful
not to touch the interior threads with fingers. Add FBS, ESC-S and LIF solution into basal
medium in a sterile field, mix well and then the reconstituted medium is ready for use.
Since several components of MESCMS are light-labile, it is recommended that the
medium not be exposed to light for lengthy periods of time. If the medium is warmed
prior to use, do not exceed 37℃. When stored in the dark at 4℃, the reconstituted
medium is stable for one month.
