藍(lán)色葡聚糖以受控的葡聚糖片段為原料，與Cibacron Blue F3GA反應(yīng)合成得到，經(jīng)非結(jié)合染料純化后，對(duì)產(chǎn)物的平均分子量、溶解度、取代度、游離染料和干燥失重進(jìn)行了控制。低分子量藍(lán)色葡聚糖（BD5、BD10和BD20）在生產(chǎn)過(guò)程中需要通過(guò)色譜法進(jìn)行純化，因此比大分子量的藍(lán)色葡聚糖更昂貴。TdB實(shí)驗(yàn)室提供各種分子量的藍(lán)色葡聚糖，范圍從5 kDa到2000 kDa。
合成與結(jié)構(gòu)
葡聚糖來(lái)自腸系膜明串珠菌B-512F，其本質(zhì)上是一個(gè)線性的α-（1-6）-連接的葡萄糖鏈，沿鏈分布的α-（1-3）支鏈百分比較低（2-5%）。所使用的葡聚糖組分的重量平均分子量（Mw）為4000到2000000。葡聚糖用Cibacron Blue F3GA標(biāo)記合成藍(lán)色葡聚糖。
產(chǎn)品由葡聚糖的近似分子量表征。BD5具有約5000的分子量。通過(guò)凝膠色譜（GPC）測(cè)定的實(shí)際分子量來(lái)確定。該值隨分析證書(shū)一起提供。

物理性質(zhì)
藍(lán)色葡聚糖是一種藍(lán)色、無(wú)味粉末，極易溶解于水或電解質(zhì)溶液。由于Cibacron Blue結(jié)構(gòu)中同時(shí)存在還酸鹽和氨基，該產(chǎn)品具有顯著的兩親性特征。由于這種特性會(huì)影響溶液的流體動(dòng)力學(xué)體積以及與凝膠的相互作用，因此在GPC檢測(cè)樣品時(shí)會(huì)發(fā)現(xiàn)一些異常。藍(lán)色葡聚糖不溶于大多數(shù)有機(jī)溶劑，例如乙醇、甲醇、丙酮、氯仿、乙酸乙酯等等。圖.2藍(lán)色葡聚糖110水溶液的吸收光譜。藍(lán)色所色團(tuán)的最大吸光度為621.5nm（見(jiàn)圖2）

穩(wěn)定性
藍(lán)色葡聚糖穩(wěn)定性尚未有前瞻性的研究。然而，對(duì)室溫下3年藍(lán)色葡聚糖樣品未顯示出任何不穩(wěn)定。建議將產(chǎn)品儲(chǔ)存在密封容器中，避光。藍(lán)色葡聚糖可在室溫下儲(chǔ)存。 藍(lán)色葡聚糖在室溫下干燥儲(chǔ)存在密封良好的容器中可穩(wěn)定6年以上
毒性
Haz-Map數(shù)據(jù)庫(kù)中藍(lán)色葡聚糖被歸為皮膚、眼睛和呼吸道刺激物。建議與大多數(shù)粉末一樣，應(yīng)避免吸入，尤其是反復(fù)接觸。
應(yīng)用
長(zhǎng)期以來(lái)藍(lán)葡聚糖2000一直用作色譜分析的空白體積標(biāo)記，藍(lán)色葡聚糖凝膠共軛物用于色譜分析也已經(jīng)許多年（參見(jiàn)GE Healthcare Life Sciences；尺寸排除色譜法；原理和方法）。現(xiàn)在TdB提供的藍(lán)色葡聚糖拓展了生物科學(xué)中的應(yīng)用范圍，且可使用標(biāo)準(zhǔn)UV和RI檢測(cè)器進(jìn)行檢測(cè)。藍(lán)葡聚糖已經(jīng)用于其他重要的研究領(lǐng)域，如下所列：溶酶體活性研究（1）、內(nèi)皮細(xì)胞通透滲透性（2）、牛精子滲透性y（3）、角膜滲透性（4）、肺血流研究（5-7）、大腦滲透性（8-10）、蛋白質(zhì)和酶與藍(lán)色葡聚糖的結(jié)合（11-13）。
